Pfizer is working on consolidating its pipeline in pain, based on some new leads and other steps to strenghten their position in the market of neuropathic pain. They are working on compounds and innovations for:
And on combination therapy for pain: Combo therapy with PF-4880682. Furthermore they are looking into new α2δ Ligands to 'Expand a Successful Class of Drugs'
Pfizer R&D pipeline in neuropathic painS,S-reboxetineS,S-reboxetine, a selective Norepinephrine Reuptake Inhibitor. S,S- reboxetine has to help fulfill the unmet need gap related to side-effects of other neuropathic analgesic drugs. Nerve growth factor mAb: PF-4383119 PF-4383119 (RN-624) for chronic low back pain, post-herpetic neuralgia and visceral pain (phase III?) PH-797804: p38 Kinase Inhibitor Inhibits Cytokine Production for post-herpetic neuralgia in phase III? Lyrica enhancement via combination therapy: PF-4880682 with Lyrica Potent, selective, antagonists of CC chemokine receptor 2 (CCR2): its efficacy in nociceptive and neuropathic models of pain comparable to celecoxib and gabapentin respectively. On research focus os on voltage-gated sodium ion channels, as these channels may play critical role in pain transduction and conduction. Program has been set up to invent selective NaV 1.7 modulators for these channels. In total Pfizer claimed last year to have 11 Compounds in Clinical Testing in the field of pain. In 2010 the following CNS drugs are mentioned by Pfizer: Phase I CNS pipeline PfizerCP-601927 Depression PF-2400013 Schizophrenia PF-2545920 Schizophrenia PF-3049423 Stroke Recovery PF-3463275 Schizophrenia PF-4455242 Bipolar Disease PF-5208769 (Vabicaserin) Schizophrenia PF-5212362 (Begacestat) Alzheimer’s Disease PF-5212377 (WYE-103760) Alzheimer’s Disease PF-4427429 Migraine PF-4457845 Pain Acute and Chronic Pain PF-4531083 Pain Severe Chronic Pain Phase II CNS pipeline PfizerChantix Cognition in Alzheimer’s Disease PF-5212365 (SAM-531) Schizophrenia, Alzheimer’s Disease PF-4383119 (tanezumab) Broad Chronic Pain (Biologic) PF-4856880 Adolor Pain PF-4856881 Adolor Chronic Pain Phase III CNS pipeline Pfizer Lyrica Alpha-2 Delta Ligand Restless Legs Syndrome and other indication extensions Latrepirdine (Dimebon) MpTp Modulator Alzheimer’s Dementia Latrepirdine (Dimebon) MpTp Modulator Huntington’s Disease Pfizer's statement on its commitment to CNSIn the U.S. today, 7 of the 10 leading causes of disability are neurological and psychiatric disorders. To meet these patient needs, Pfizer is taking a bold leadership approach that will evolve from dealing with symptoms to modifying diseases, where scientifically feasible. As a result, Pfizer has new approaches to attack Alzheimer’s disease, schizophrenia and other debilitating conditions. Pfizer's statement related to painIn recent years there have been very few breakthrough medicines for the treatment of chronic pain, with many of the current standard medicines dating back decades (if not centuries). The result of this is that there are still millions of patients across the world for whom there is no adequate medicine to treat their pain. The impact of pain is profound. Frequent sufferers are stopped from working, sleeping, and socializing, with a common result of depression. Pfizer’s pain team hopes that new insights into the causes of pain, and new ways to actually ‘measure’ it, can help usher in a series of new medicines that will provide greater relief from the symptoms of pain, allowing patients to return to normal lives at work and at home. From the Centre for the study and treatment of neuropathic pain and neuropathy in Soest, the NertherlandsThis site helps patients and treating physicians, neurologists, anesthesiologists and other pain specialists to find the best and most up to date research findings related to neuropathy and neuropathic pain and the treatment thereof. In our centre we are specialised in treating patients suffering from neuropathic pain and neuropathy following an Integrated Medicine concept. Part of our activities are within the field of consultation. We assist pharmaceutical companies in R&D strategies related to finding new drugs to treat neuropathic pain and neuropathy. March 2010, Jan M. Keppel Hesselink, MD, PhD
